Roxana joined Phenomenex in 2018 as an Applications Scientist and is currently working with the applications team to produce analytical solutions for the biopharma industry. Her current work focuses on characterization of mRNA, sgRNA and siRNA. Prior to Phenomenex, Roxana worked for Caltech and the University of Southern California developing LM-CS workflows for the proteomics community. She received her bachelor’s and master’s degree in Chemistry and Biochemistry from California State University, Los Angeles.

Overview
Antibody–oligonucleotide conjugates (AOCs) combine antibody specificity with oligonucleotide precision, offering a promising therapeutic platform. Accurate oligo-to-antibody ratio (OAR) determination is essential for efficacy and safety, yet conventional hydrophobic interaction chromatography (HIC) fails due to the hydrophilic, polyanionic nature of oligonucleotides, which masks hydrophobic differences. Native size-exclusion chromatography coupled with mass spectrometry (SEC-MS) has emerged as a robust approach for intact AOC analysis, enabling OAR distribution and aggregate detection.  This webinar will showcase an SEC-MS method developed to characterize AOCs composed of a trastuzumab biosimilar conjugated with siRNA.
